Molecular mechanism of non-enzymic glication of proteins and meaning of this process in diabetes

Hyperglycemia plays a important role in diabetes angiopathy mechanisms. It causes damage through activation a several metabolic pathways. One of them is non-enzymic glication of proteins. This is non-enzymic association of sugars and proteins, which have large amount of free amino groups. In effect of this reaction originate advanced glication end products (AGEs). They are stable, with bronze colour, specific properties and they have capacity for making cross-links between proteins. They have specific for themselves receptors on a few types of cells membranes. Most important proteins which are bound by non-enzymic glication are: hemoglobin, albumin, collagen and crystallin. Glycated hemoglobin and glycated albumin indication is very important in hyperglycemia diagnosis. Glication of collagen and other proteins in matrix causes in diabetes stiffnes in vessel walls: in veins and also in arteries, whereas crystallin glication in eye lens causes cataract. Investigation about invention of a drug which will be preventing from non-enzymic glication still continue, but they have many methodic difficulties. Creation of this kind of medicine will be crucially for diabetes therapy.
