Ketoprofen as analgesic drug with effective anti-inflammatory effect in rheumatic diseases

Pain is the first and essential symptom of most rheumatic diseases. Ketoprofen is a non-steroidal anti-inflammatory drug (NSAID), with strong anti-inflammatory, analgesic, and anti-pyretic effect. The main mechanism of action of ketoprofen is inhibition of cyclooxygenase (COX), which results in inhibition of prostanoid synthesis (prostaglandins, prostacyclin, thromboxane). Moreover, ketoprofen inhibits bradykinin activity and inhibits lipo-oxygenase, which results in inhibition of leukotriene synthesis. Ketoprofen may be used as a symptomatic treatment in different types of arthritis, osteoarthritis, and soft tissue rheumatism, usually if pain exacerbates. Results of clinical trials indicate that the effects of ketoprofen therapy are better than or comparable to other NSAIDs. The safety profile is also similar to other NSAIDs; however, it is recommended to assess the individual risk of adverse events before treatment.
